Okay, so I actually think this is a stupid statement.
Any band worth a damn has a guitarist capable capturing vote for such a poll.
In fact a simple youtube search will yield amateurs imitating their favorite guitarists and doing an excellent job of it.
But, this place is deader than a door-nail, so maybe this will illicit a few replies providing a little entertainment.

Maybe instead of their playing ability, which they all have, the criteria should be their ability to write a catchy lick?

Marty Friedmans Megadeth stint.(Especially rust in peace album)

I think to narrow the field you would have to look at it specific terms Did they have an effect on how people actually play the guitar or the sounds that came from the guitar.

Jimi Hendrix - impact on how people actually played and linked notes together on the guitar

Tom Scholz - developed sounds produced with the guitar

Zakk Wylde is a shredder, Prince (not really a fan) was also an absolute shredder that doesn't get enough credit for how good he was. Kirk Hammett deserves some love too

No order, just who comes to mind:
Jimi, Prince, Eddie, Clapton, Vaughan, Santana, BB King, Chuck Berry, Jimmy Page, Johnny Ramon, Kirk Hammett. I love Tom Morello, but He's not in the greatest conversation. Slash is solid, but not greatest. Also a big Mark Knopfler fan.

If I had to pick ONE, I'd personally say Jimi or Prince.
